There have been no previous reports on the influence of cryopreserved ensheathing cells on axonal regeneration. To observe whether cryopreserved olfactory ensheathing cells could be used to promote axonal regeneration in a spontaneously non-regenerating system, cryopreserved ensheathing cells were transplanted into a transected spinal cord.

In primary cultures of the olfactory bulb (OB) of adult rat, ensheathing cells were purified away from other cell types after 1 week culture. The cells were stored at -80ºC for 1 month before transplantation. After laminectomy at the lower thoracic level, the spinal cords of adult rats were exposed and completely transected at T10. A suspension of ensheathing cells were injected into the lesion site in 12 adult rats, and control D/F-12 (1:1 mixture of DMEM and Ham’s F-12) were injected in 8 adult rats.

Two weeks and eight weeks after cells transplantation, numerous regenerating axons were observed that grew through the transplant and continued to regenerate into the denervated host tract. Cell labelling using anti-Myelin Basic Protein (MBP) and anti-Nerve Growth Factor Receptor (anti-NGFR) indicated that the regenerated axons were derived from the appropriate neural source, and the donor cells migrated into the denervated host tract. But axonal degeneration existed and regenerating axons was not observed within the spinal cord of adult rats with only D/F-12 injection.

In conclusion, cryopreserved ensheathing cells transplantation can be used to promote axonal regeneration in transected adult rat spinal cord, and provide a useful OECs source for future clinical trials.
