THE FATE OF GRAFTED NEURAL STEM CELLS IN THE SPINAL CORD FOLLOWING SPINAL ROOT AVULSION Imprimer Envoyer

Objective: Avulsion of spinal root results in death of motoneurons in corresponding spinal segment. In order to explore whether the grafted neural stem cells (NSCs) could replace the dead motoneurons, the green fluorescent protein (GFP) positive NSCs were transplanted into the ventral horn of the spinal cord after root avulsion.

Methodology: The NSCs were isolated from the hippocampus of postnatal GFP transgene rats, cultured in the serum free media with bFGF and EGF, and identified by the nestin –immunocytochemistry. Four weeks after the avulsion, the GFP-positive NSCs were microinjected into the ventral horn of the spinal cord, and the avulsed ventral root was reimplanted onto the spinal cord.

Result: 6 or 8 weeks after transplantation, the GFP-positive cells (grafted cells) mainly congregated around the injected site, their bodies’ size was about 10um diameter. No GFP cells were labeled by fluoroGold which was microinjected into the reimplanted nerve 2 days before sacrifice. The immunostainings showed that, most the GFP cells were nestin positive, less of them were neurofilament positive or GFAP positive, and no of them MAP2 positive.

Conclusion: The findings indicated that the majority of grafted NSCs in the ventral horn of the spinal cord mainly maintained undifferentiated state and difficult replace the dead motoneurons due to root avulsion.
