Objective: To investigate the cellular properties, immunologic characters and transplantation of human foetal olfactory ensheathing cells (OECs) in spinal cord injury.

Methods: Human foetal OECs were obtained from olfactory bulbs and bone marrow stem cells (MSCs) were isolated from human bone marrow aspiration, stained with fluorescein isothiocyanate (FITC)–conjugated or phycoerythrin (PE)–conjugated monoclonal antibodies, and analyzed using a FACScan laser flow cytometry system. Lymphocytes were mixed at a 1:1 ratio with OECs/MSCs in MLR. Lymphocytes proliferation was measured on day 6 as mean 3H-TdR uptake in counts per minute (c.p.m.). Coculture of cortical neurons/OECs to test for the influence of OECs on neurite growth.

Results: Both OECs and MSCs are negative for HLA-DR, B7-1(CD80), B7-2(CD86), CD40, CD40L, histocompatibility antigens related to GVHD or tolerogenic activities. OECs and MSCs could weakly stimulate the proliferation of lymphocytes in MLR compared with the control MLR, but Student t test showed no significant difference (p>0.05).

Conclusion: The study suggested that human foetal OECs are not significantly immunogenic.
